目的:建立大黄饮片MEKC-DAD指纹图谱分析方法,并对大黄及其炮制品的指纹谱进行比较。方法:选择胶束电动毛细管电泳分离模式,以25 mmol.L-1硼砂-25 mmol.L-1SDS-10%乙腈(pH10.90)为运行缓冲液,分离电压12 kV,以大黄酸为参照物(IS),测定其指纹图谱,并作模糊聚类法分析和相似度评价。结果:初步建立了以11个共有峰为特征指纹信息的10批大黄地产药材MEKC-DAD指纹图谱;发现少数产地大黄药材的指纹图谱有一定差异,生品与其炮制品的指纹谱中共有峰相对峰面积差异显著。结论:方法准确可靠,重复性好,可作为大黄饮片内在质量评价的依据。
Objective: To establish the method of MEKC-DAD fingerprint analysis of rhubarb decoction and to compare the fingerprints of rhubarb and its processed products. Methods: Micellar electrokinetic capillary electrophoresis separation mode was set up. With 25 mmol.L-1 borax-25 mmol.L-1SDS-10% acetonitrile (pH10.90) as the running buffer, the voltage was 12 kV and Rhein was used as a reference (IS). The fingerprints were determined and analyzed by fuzzy clustering and similarity evaluation. Results: MEKC-DAD fingerprinting of 10 batches of rhubarb rhizoma herbs was established preliminarily. Conclusion: The method is accurate and reliable with good repeatability and can be used as the basis for evaluating the intrinsic quality of Rhubarb Pieces.
