目的检测中国汉族急性白血病(AL)患者和健康受试者中CYP3A4*18和CYP3A5*3基因型分布特征,探讨CYP3A4和CYP3A5基因多态性与中国汉族AL易感性的相关性。方法病例对照研究,用聚合酶链反应(PCR)后直接测序的方法,检测中国汉族AL患者91例,健康受试者200例的CYP3A4*18B和CYP3A5*3位点的基因型分布及等位基因频率。结果中国汉族AL患者和健康受试者CYP3A4*18B和CYP3A5*3基因型分布均符合Hardy-Weinberg平衡。与健康受试者比较,AL患者CYP3A4*18B基因型及等位基因频率差异均无统计学意义(P>0.05),CYP3A5*3基因型及等位基因分布频率差异均有统计学意义(P<0.05)。结论 CYP3A4*18B多态性与中国汉族AL的易感性可能无关,CYP3A5*3多态性与中国汉族AL的易感性可能相关。
Objective To detect the distribution of CYP3A4 * 18 and CYP3A5 * 3 genotypes in Chinese Han patients with acute leukemia (AL) and healthy subjects and to explore the relationship between CYP3A4 and CYP3A5 polymorphisms and AL susceptibility in Han Chinese. Methods A case-control study was conducted to detect the genotypes and alleles of CYP3A4 * 18B and CYP3A5 * 3 loci in 91 Chinese AL patients and 200 healthy subjects by polymerase chain reaction (PCR) Gene frequency. Results The distribution of CYP3A4 * 18B and CYP3A5 * 3 genotypes in AL patients and healthy subjects in Han Chinese were in accordance with Hardy-Weinberg equilibrium. Compared with healthy subjects, there was no significant difference in CYP3A4 * 18B genotypes and allele frequencies between patients with AL (P> 0.05), CYP3A5 * 3 genotypes and allele frequencies (P <0.05). Conclusion The CYP3A4 * 18B polymorphism may not be related to AL susceptibility in Chinese Han population. CYP3A5 * 3 polymorphism may be associated with AL susceptibility in Chinese Han population.
