目的:探讨胸腺肽对慢性阻塞性肺病(COPD)患者树突状细胞的免疫调节作用。方法:分离COPD患者和健康者外周血单个核细胞并诱导成树突状细胞,在体外通过胸腺肽干预树突状细胞分为四组,分别为COPD胸腺肽刺激组、COPD组、健康者胸腺肽刺激组、健康组,通过ELISA方法检测各组外周血树突状细胞分泌Th1型细胞因子IFN-γ和Th2型细胞因子IL-4的表达水平。结果:COPD胸腺肽刺激组较COPD组和健康者胸腺肽刺激组IFN-γ明显升高(分别P<0.05和P<0.01),而COPD胸腺肽刺激组较COPD组IL-4的表达明显降低(P<0.05),COPD组较健康组分泌细胞因子也明显增多(P<0.01)。结论:胸腺肽可调节COPD患者IFN-γ、IL-4的分泌表达,有利于改善COPD患者的免疫功能。
Objective: To investigate the immunomodulatory effect of thymosin on dendritic cells in patients with chronic obstructive pulmonary disease (COPD). Methods: Peripheral blood mononuclear cells (PBMCs) from patients with COPD and healthy individuals were isolated and induced into dendritic cells. Thymocyte-mediated dendritic cells (DCs) were divided into four groups: COPD thymosin group, COPD group, healthy thymosin group , Healthy group. The levels of Th1-type cytokine IFN-γ and Th2-type cytokine IL-4 secreted by peripheral blood dendritic cells in each group were detected by ELISA. Results: Compared with COPD group and healthy thymosin group, the IFN-γlevels in COPD group were significantly increased (P <0.05 and P <0.01, respectively), while the expression of IL-4 in COPD thymosin group was significantly lower than that in COPD group (P < 0.05). The levels of cytokines in COPD group were significantly higher than those in healthy group (P <0.01). Conclusion: Thymosin can regulate the secretion of IFN-γ and IL-4 in patients with COPD, which is helpful to improve the immune function of COPD patients.
