应用NorthernBlot和DotBlot杂交技术研究了青春期糖尿病时大鼠附睾 5α 还原酶基因表达的变化。实验分对照 (C)组、糖尿病 (D)组和胰岛素治疗糖尿病 (ID)组。结果 :NorthernBlot方法显示附睾头部 5α 还原酶表达强度 ,D组较C组显著降低 (P <0 .0 1) ,ID组较D组显著升高 (P <0 .0 0 1) ,体、尾部各组间变化不显著 (P >0 .0 5 ) ;DotBlot结果 ,除头部ID组未见恢复和体部D组显著高于C组外 ,其余结果与Northernblot基本一致。结果提示 :青春期糖尿病时可能由于糖尿病状态引起附睾 5α 还原酶表达降低 ,胰岛素治疗后随糖尿病状态改善 ,附睾头部 5α 还原酶表达升高
Northern blot and DotBlot hybridization were used to study the changes of rat epididymal 5α-reductase gene expression during adolescent diabetes. The experiment was divided into group (C), diabetes (D) and insulin-treated diabetic (ID) groups. Results: NorthernBlot showed that the expression of 5α-reductase in the head of the epididymis was significantly lower than that in the C group (P <0.01), the ID group was significantly higher than the D group (P <0.01) There was no significant difference between the tail groups (P> 0.05). The results of DotBlot were similar to those of Northern blot except that the head ID group did not recover and the body group D was significantly higher than that of the C group. The results suggest that the expression of 5α-reductase in epididymis may be decreased due to the state of diabetes during adolescent diabetes, and the expression of 5α-reductase in the epididymis head may be increased with the improvement of diabetic state after insulin treatment
