目的:建立同时测定盾叶薯蓣根中黄姜素A、盾叶新苷及薯蓣皂苷3种成分含量的HPLC-ELSD分析方法,考察不同产地盾叶薯蓣根中3种成分的含量。方法:药材粉末经70%乙醇回流提取,以HPLC-ELSD法进行测定。色谱柱采用Diamosil C18(250mm×4.6mm,5μm),流动相为甲醇(A)-水(B),梯度洗脱(0~10min,78%A;10~13min,78%A→90%A;13~45min,90%A),流速为1.0mL·min-1,漂移管温度为79.0℃,气体流速为2.1L·min-1。结果:黄姜素A、盾叶新苷和薯蓣皂苷三者均有良好的线性关系;其平均回收率(n=6)分别为100.4%,103.0%,95.3%。结论:本方法操作简便,结果可靠,可用于同时定量分析盾叶薯蓣根中黄姜素A、盾叶新苷及薯蓣皂苷3种成分。
OBJECTIVE: To establish a HPLC-ELSD method for the simultaneous determination of three constituents of Vetiveria zizanioides, Aegilops folium and Dioscin in Dioscorea zingiberensis CHWright, and to study the content of three constituents in Dioscorea zingiberensis from different habitats. Methods: The medicinal powder was extracted by 70% ethanol and determined by HPLC-ELSD. The column was eluted with Diamosil C18 (250 mm × 4.6 mm, 5 μm) and the mobile phase consisted of methanol (A) -water (B), gradient elution (0-10 min, 78% A; 10-13 min, 78% A → 90% A ; 13-45min, 90% A), the flow rate was 1.0mL · min-1, the drift tube temperature was 79.0 ℃ and the gas flow rate was 2.1L · min-1. Results: The results showed that there was a good linear relationship between the three compounds of yingcin A, sanguinoside and diosgenin. The average recoveries (n = 6) were 100.4%, 103.0% and 95.3%, respectively. Conclusion: The method is simple and reliable, and can be used for the simultaneous quantitative analysis of three constituents of yacon, shield leaf and diosgenin in Dioscorea zingiberensis.
