目的:制备细胞因子激活杀伤(CIK)细胞并观察其生物学特性及对不同肿瘤细胞株的杀伤作用。方法:外周血单个核细胞用无血清培养基经干扰素γ(IFNγ),CD3McAb,白介素2(IL2),白介素1α(IL1α)诱导,制作CIK细胞,以淋巴因子激活的杀伤细胞(LAK)作为对照,流式细胞仪检测CIK细胞的免疫表型,MTT法检测其对不同肿瘤细胞株的杀瘤活性。结果:CIK细胞与LAK细胞相比增殖速度快、最终增殖倍数高。CIK细胞主要由CD3和CD56双阳性细胞构成。CIK细胞对多种肿瘤细胞具有较强的杀伤活性。结论:CIK细胞过继免疫治疗是一种非常有前景的抗肿瘤治疗方法。
OBJECTIVE: To prepare cytokine-activated killer (CIK) cells and observe their biological characteristics and killing effects on different tumor cell lines. Methods: Peripheral blood mononuclear cells were induced by IFNγ, CD3McAb, IL2 and IL1α in serum-free medium. Lymphokine-activated killer cells (LAK) were used as The immunophenotypes of CIK cells were detected by flow cytometry. The killing activity of CIK cells on different tumor cell lines was tested by MTT assay. Results: Compared with LAK cells, CIK cells proliferated rapidly and their final multiplication rate was high. CIK cells are mainly composed of CD3 and CD56 double positive cells. CIK cells on a variety of tumor cells with strong killing activity. Conclusion: CIK cell adoptive immunotherapy is a very promising anti-tumor therapy.
