增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)是一种分子量3.6×10~4的核蛋白,这种蛋白与细胞周期和增殖细胞 G_1晚期和 S 期核内聚积有关。在各种恶性肿瘤中,肿瘤细胞的增殖活性与肿瘤的分级、转移潜力、复发和预后相关,已被作为识别细胞周期 G_1/S 期内源性标志物。国内有关PCN A 在人原发性肝癌(PHC)中研究报道很少。我们用单克隆抗体 PC10检测50例 PHC 及其癌旁肝组织中 PCNA 表达分布,旨在探讨细胞增殖活性与 PHC 临床病理特征的关系及其预后意义。资料与方法一、资料收集手术切除病理诊断明确的 PHC 标本50例。每例取癌及癌旁肝组织各一块,同时收集非肝病死亡的尸检肝组织10例作正常对照,组织经10%甲醛固定,石蜡包埋,切片,分别进行 HE 和免疫组织化学染色。50例 PHC 中,肝细胞性肝癌
Proliferating cell nuclear antigen (PCNA) is a nuclear protein with a molecular weight of 3.6×10~4. This protein is involved in the nuclear accumulation in the cell cycle and proliferating cells G_1 late and S phase. In various malignancies, the proliferative activity of tumor cells correlates with tumor grade, metastatic potential, recurrence, and prognosis, and has been used as an endogenous marker to identify G1/S phase in the cell cycle. Domestic studies on PCN A in human primary liver cancer (PHC) are rare. We used monoclonal antibody PC10 to detect the expression of PCNA in 50 cases of PHC and its paraneoplastic liver tissues in order to investigate the relationship between cell proliferation activity and the clinicopathological features of PHC and its prognostic significance. Materials and methods 1. Data collection Surgical resection of 50 specimens of pathologically confirmed PHC specimens was performed. One case of cancer and one adjacent liver tissue were collected, and 10 non-liver disease autopsied liver tissues were collected as normal controls.
