目的探索胞内钙离子在Nogo-A抑制轴突生长中的作用,以进一步了解Nogo-A抑制轴突生长的信号转导机制。方法利用激光共聚焦显微镜技术,检测Nogo-A作用于小脑颗粒细胞后不同时段胞内钙离子的浓度,并观察加入钙通道阻滞剂———尼莫地平对Nogo-A抑制轴突生长的影响。结果小脑颗粒细胞内钙离子浓度升高发生于加入Nogo-A后5 min,30 min时达到最高,然后逐渐下降,2 h后恢复至正常水平;加入尼莫地平组的轴突生长与对照组相比有显著差异。结论钙离子可能参与了Nogo-A抑制轴突生长的信号转导。
