目的研究银杏外种皮提取物有效部位GBEE-2体外对人胃癌细胞凋亡及其凋亡通路的影响。方法体外培养人胃癌SGC-7901细胞,应用MTT法检测细胞增殖;应用流式细胞术检测凋亡率和细胞周期;应用ELISA法检测培养上清中基因蛋白Caspase-9、Fas及Survivin的含量。结果 GBEE-2(5~160μg.mL-1)对SGC-7901细胞的体外增殖具有抑制作用,并具有浓度依耐性,IC50为83μg.mL-1;在10~160μg.mL-1浓度下体外培养,GBEE-2可提高SGC-7901细胞的凋亡率,可使Caspase-9、Fas蛋白表达量增加(P<0.05或0.01),而Survivin蛋白的表达则减少(P<0.05或0.01)。结论 GBEE-2抑制人胃癌SGC-7901细胞的作用机制与诱导细胞凋亡有关,其凋亡通路涉及Caspase途径。
Objective To study the effect of gibexter ginkgo biloba extract (GBEE-2) on apoptosis and apoptosis pathway in human gastric cancer cells in vitro. Methods Human gastric cancer SGC-7901 cells were cultured in vitro. Cell proliferation was detected by MTT assay. The apoptosis rate and cell cycle were detected by flow cytometry. The levels of Caspase-9, Fas and Survivin in culture supernatant were detected by ELISA. Results GBEE-2 (5 ~ 160μg.mL-1) had inhibitory effect on the proliferation of SGC-7901 cells in vitro with IC50 of 83μg.mL-1 at concentrations of 10 ~ 160μg.mL-1 GBEE-2 could increase the apoptosis rate of SGC-7901 cells and increase the expression of Caspase-9 and Fas protein (P <0.05 or 0.01), while the expression of Survivin protein decreased (P <0.05 or 0.01). Conclusion The mechanism of GBEE-2 inhibiting human gastric cancer SGC-7901 cells is related to the induction of apoptosis, and the apoptosis pathway involves Caspase pathway.
