目的:观察红背丝绸2种提取物和单体(CA1201)对内皮素1(endothelin1,ET1)生物效应的拮抗作用。方法:用红背丝绸3种提取物对ET1致小鼠急性死亡、血管收缩和升压作用的拮抗,探索用中草药拮抗ET的生物效应。结果:3种提取物对ET1致小鼠急性死亡有明显的保护作用,使小鼠的死亡率下降;并可拮抗ET1致胸主动脉的缩血管效应,醇提物和乙酸乙酯提取物(10-7母液)拮抗ET缩血管效应分别为57.27%和58.61%;CA120110-8mol·L-1和10-9mol·L-1亦能拮抗ET1的缩血管效应;醇提物和乙酸乙酯提取物ig及CA1201iv后能拮抗ET1的升压作用,拮抗ET1作用均呈现明显的剂量依赖性。结论:红背丝绸2种提取物和CA1201可拮抗ET1的生物学效应
Objective: To observe the antagonistic effects of two kinds of extracts and monomers (CA1201) of red back silk on the biological effects of endothelin1 (ET1). METHODS: Three kinds of extracts from red-backed silk were used to antagonize acute death, vasoconstriction and pressure increase induced by ET1 in mice. The use of Chinese herbal medicines was explored to antagonize the biological effects of ET. RESULTS: The three extracts had obvious protective effects on acute death of mice induced by ET1, which reduced the mortality rate of mice, and could antagonize the vasoconstrictor effect of thoracic aorta induced by ET1. The antagonism of ET vasoactive effects by ester extracts (10-7 mother liquor) was 57.27% and 58.61%, respectively. CA120110-8mol·L-1 and 10-9mol·L-1 also antagonized ET1. The vasoconstrictive effect; ethanol extract and ethyl acetate extract ig and CA 1201iv can antagonize the vasopressor action of ET 1 and antagonize the effects of ET 1 in a dose-dependent manner. Conclusion: Two kinds of extracts of red back silk and CA1201 can antagonize the biological effects of ET1
