目的:探讨抵当汤对慢性前列腺炎大鼠的抗炎作用及调节免疫机制。方法:将50只大鼠随机分为假手术组(蒸馏水10 mL.kg-1)、病理模型组(蒸馏水10 mL.kg-1)、抵当汤高剂量组(16.2 g.kg-1)、抵当汤低剂量组(8.1 g.kg-1)、阳性对照组(前列康片0.5 g.kg-1)。在大鼠前列腺双侧背叶分别注入消痔灵注射液0.1 mL,5周后形成慢性前列腺炎;假手术组于同样部位同样方法注入0.9%氯化钠注射液0.1 mL。各组大鼠于术后开始ig给药,假手术组及病理模型组ig蒸馏水,连续给药5周后,检测前列腺组织匀浆中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、免疫球蛋白G(IgG)。病理形态学观察前列腺组织腺体管腔、分泌物、间质炎细胞、纤维组织。结果:与病理模型组大鼠前列腺组织匀浆中TNF-α,IL-6 IgG(67.63±15.15),(56.43±23.75),(23.26±2.57)ng.L-1比较,其他4组均较低,差异均具有显著性(P<0.05);与阳性药物组大鼠前列腺组织匀浆中TNF-α,IgG(37.90±13.64),(19.08±2.47)ng.L-1比较,抵当汤高剂量组大鼠TNF-α,IgG(23.88±8.18),(15.94±1.24)ng.L-1含量较低,差异均具有显著性(P<0.05);与抵当汤低剂量组大鼠前列腺组织匀浆中TNF-α(39.80±11.12),IgG(19.22±1.11)ng.L-1含量比较,抵当汤高剂量组TNF-α,IgG含量较低,差异均具有显著性(P<0.05)。结论:抵当汤降低了大鼠前列腺组织匀浆中TNF-α,IL-6,IgG含量,从而改善了免疫功能,抑制了炎症反应。
Objective: To investigate the anti-inflammatory effect and the regulatory mechanism of Daodang Decoction in rats with chronic prostatitis. Methods: Fifty rats were randomly divided into sham operation group (distilled water 10 mL.kg-1), pathological model group (distilled water 10 mL.kg-1), high-dose group of dengtang decoction (16.2 g.kg-1) Dangdang Tang low-dose group (8.1 g.kg-1), positive control group (Qianlie Kang 0.5 g.kg-1). 0.1 ml of Xiaozhiling injection was injected into both sides of the prostate of rats respectively, and chronic prostatitis was formed after 5 weeks. In the sham-operated group, 0.1 mL of 0.9% sodium chloride injection was injected in the same way. Rats in each group were given ig administration after operation. Rats in sham operation group and pathological model group were treated with distilled water. After continuous administration for 5 weeks, the levels of tumor necrosis factor-α (TNF-α), interleukin- 6 (IL-6), immunoglobulin G (IgG). Pathological observation of prostate gland duct, secretions, interstitial cells, fibrous tissue. Results: Compared with the pathological model group, the levels of TNF-α, IL-6 IgG (67.63 ± 15.15), (56.43 ± 23.75), (23.26 ± 2.57) ng.L-1 in the prostate tissue homogenate were significantly higher in the other four groups (P <0.05). Compared with the positive drug group, the levels of TNF-α, IgG (37.90 ± 13.64) and (19.08 ± 2.47) ng.L- The levels of TNF-α, IgG (23.88 ± 8.18) and (15.94 ± 1.24) ng.L-1 in the dose group were significantly lower than those in the low dose group (P <0.05) The levels of TNF-α and IgG in high dose group of Dendang Decoction were lower than those in the control group (P <0.05), and the difference was significant (P <0.05) . Conclusion: DaoTang decoction can reduce the content of TNF-α, IL-6 and IgG in the homogenate of rat prostatic tissue, thus improving the immune function and inhibiting the inflammatory reaction.
