目的比较加味当归补血颗粒(JW)与当归补血汤(DG)对环磷酰胺(CTX)所致小鼠的骨髓抑制及抑瘤作用,观察JW对荷瘤小鼠红细胞免疫功能的影响。方法清洁级KM小鼠随机分为CTX组、荷瘤组及正常对照组。CTX组腹腔注射CTX,同时灌胃给予JW或DG,10d后检测动物外周血象:白细胞、红细胞、血小板及骨髓有核细胞数,进行组间比较;荷瘤组常规接种H22(小鼠肝癌)造模后,灌胃给予JW,10d后检测小鼠红细胞免疫功能、红细胞免疫复合物花环率(RBC-ICRR)及红细胞C3b受体花环率(RBC-C3BRR),同时测定对CTX抑瘤作用的影响。结果①CTX给药后小鼠外周血象及骨髓有核细胞数均有显著性下降,给予JW后各检测值的下降幅度均小于CTX组,与正常对照组比较差异无显著性。DG的作用较JW小。②JW高剂量组对荷瘤小鼠红细胞免疫功能具有显著的增强作用。③JW或DG与CTX合用,抑瘤率较单用CTX均有一定的提高,但与CTX组比较差异无显著性。结论加味当归补血颗粒具有抑制CTX毒性的作用,可明显提高荷瘤小鼠的红细胞免疫功能,与CTX合用能起到一定的抑瘤增效作用。
Objective To compare the bone marrow suppression and antitumor effects of Danggui Buxue Granules (JW) and Danggui Buxue Decoction (DG) on mice induced by cyclophosphamide (CTX), and observe the effects of JW on red blood cell immune function in tumor-bearing mice. Methods Clean grade KM mice were randomly divided into CTX group, tumor-bearing group and normal control group. The CTX group was intraperitoneally injected with CTX and intragastrically administered JW or DG. After 10 days, the peripheral blood of the animals was measured: the number of white blood cells, red blood cells, platelets and bone marrow nucleated cells. The groups were compared; the tumor group was routinely inoculated with H22 (mice liver cancer). After immunization, JW was intragastrically administered, and 10 days later, the red blood cell immune function, red blood cell immune complex rosette rate (RBC-ICRR) and red blood cell C3b receptor rosette rate (RBC-C3BRR) were measured, and the effect of CTX on tumor inhibition was measured. . Results 1 The number of peripheral blood and bone marrow nucleated cells decreased significantly after CTX administration. The decrease of each test value after JW administration was smaller than that of CTX group. There was no significant difference compared with the normal control group. The role of DG is smaller than JW. 2JW high-dose group had significant enhancement of red cell immune function in tumor-bearing mice. The combination of 3JW or DG with CTX had a certain increase in tumor inhibition rate compared with CTX alone, but there was no significant difference compared with CTX group. [Conclusion] Modified Danggui Buxue Granule can inhibit the toxicity of CTX, which can significantly increase the red blood cell immune function of tumor-bearing mice. Combined with CTX can play a certain role in inhibiting tumor growth.
