流式微珠阵列法检测结脑患者Th1/Th2细胞因子表达

目的:运用流式微珠阵列法评价IFN-γ、IL-4在结核性脑膜炎(结脑)中的表达情况,以探讨Th1/Th2平衡状态与其疾病发生发展的关系。方法:采集结脑患者治疗前后的外周血和脑脊液以及健康人的外周血,流式微珠阵列法检测其中IL-4和IFN-γ的浓度,比较各检测指标在各组中差异,并分析差异的意义。结果:结脑患者外周血的IFN-γ浓度较正常对照组低,IL-4浓度无差异;治疗前后外周血中的IFN-γ、IL-4和脑脊液中的IL-4水平均差异无统计学意义,但治疗前后脑脊液中IFN-γ表达差异有统计学意义。结论:与正常对照组及治疗前后相比较,Th1细胞因子IFN-γ在结脑患者的体液中有显著性变化,提示其参与结脑的病理生理过程;流式微珠阵列法能准确检测Th1/Th2细胞因子表达的动态变化。 OBJECTIVE: To evaluate the expression of IFN-γ and IL-4 in tuberculous meningitis (tuberculous meningitis) by flow bead-array method to explore the relationship between the Th1 / Th2 balance and the occurrence and development of the disease. Methods: Peripheral blood and cerebrospinal fluid in peripheral blood and cerebrospinal fluid (CSF) of healthy people and peripheral blood of healthy people were collected before and after treatment. The concentration of IL-4 and IFN-γ in each group were measured by flow cytometry. The differences among the groups were compared and analyzed Meaning. Results: The concentration of IFN-γ in the peripheral blood of patients with tuberculous meningitis was lower than that of the normal control group, and there was no difference in the concentration of IL-4. The levels of IFN-γ, IL-4 in the peripheral blood and IL-4 in cerebrospinal fluid before and after treatment showed no statistical difference Significance, but before and after treatment of cerebrospinal fluid IFN-γ expression difference was statistically significant. Conclusion: Compared with the normal control group and before and after treatment, the expression of Th1 cytokine IFN-γ in body fluid of patients with tuberculous meningitis has significant changes, suggesting that it participates in the pathophysiology of tuberculous meningitis. The flow cytometry can accurately detect Th1 / Dynamic Changes of Th2 Cytokines Expression.
