目的用改良标准加入法替代标准曲线法对石墨炉原子吸收光谱法测定血中铅浓度的标准方法进行改进。方法全血样品经5%硝酸溶液处理,用石墨炉原子吸收法测定,改良标准加入法(以第1待测血样的基体为默认基体,绘制标准校正曲线并测定后续样品)进行定量。结果该方法在0~60μg/L范围内,回归方程为Y=0.005 2 X+0.008 5,r=0.999 1,最低检出浓度为1.20μg/L,批内精密度相对标准偏差为1.30%~3.85%,批间精密度相对标准偏差为0.67%~6.42%,加标回收率为94.01%~98.42%。结论该方法以待测血样基体为默认基体,用改良标准加入法测定,免除了标准工作曲线法中不能完全消除基体血液中本底铅所带来的干扰以及传统标准加入法每样一标准校正曲线的困扰。该方法简便快速,成本低、干扰少。
Objective To improve the standard method of determination of lead in blood by graphite furnace atomic absorption spectrometry with a modified standard addition method instead of the standard curve method. Methods The whole blood samples were treated with 5% nitric acid solution, determined by graphite furnace atomic absorption spectrometry and modified by standard addition method (the first matrix of the blood sample was taken as the default matrix, the standard calibration curve was drawn and the subsequent samples were determined). The relative standard deviations of inter-batch precision were 0.67% ~ 6.42%, and the recovery rates were 94.01% ~ 98.42%. Conclusions This method takes the blood sample matrix as the default matrix and uses the modified standard addition method to eliminate the interference caused by the standard working curve method that can not completely eliminate the background lead in the matrix blood and the standard correction method Curious curve.
