目的:研究心舒汤对模型大鼠、小鼠心律失常和心肌缺血的保护作用。方法:100只小鼠随机分成舒心汤高、中、低剂量组(1.7,0.85,0.42 g.kg-1),模型组(0.2 mL纯水)和冠心丹参胶囊组(0.35 g.kg-1),连续ig给药14 d后,复制三氯甲烷小鼠心律失常模型观察小鼠室颤发生率。大鼠50只随机分成5组,舒心汤高、中、低剂量组(1.18,0.59,0.29 g.kg-1),模型组(2 mL纯水)和冠心丹参胶囊组(0.24 g.kg-1)连续ig给药14 d,建立乌头碱致大鼠心律失常的模型,观察不同剂量舒心汤对模型动物心律失常的保护作用。60只大鼠随机分成舒心汤高、中、低剂量组,空白对照组、冠心丹参胶囊组和模型组,给药剂量、方法同上大鼠,空白对照组给药同模型组,连续给药14 d;12 d开始使用异丙肾上腺素建立大鼠心肌缺血模型,连续造模2 d,观察心舒汤对心肌缺血的保护作用。结果:心舒汤高剂量组使三氯甲烷导致的小鼠心室纤颤(VF)的发生率降低至25%;能增加导致的大鼠室性早搏(VP)、室性心动过速(VT)、室颤(VF)的乌头碱用量19.7,23.7,32.6μg.kg-1;减小ECG中T波位移幅度;使心肌缺血大鼠血清中肌酸激酶(CK)和谷草转氨酶(AST)含量降低到2 572.9,111.3 U.L-1。结论:心舒汤对心律异常和心肌缺血具有保护作用。
Objective: To study the protective effect of Xinshu decoction on arrhythmia and myocardial ischemia in model rats and mice. Methods: 100 mice were randomly divided into Shuxin Tang high, medium and low dose groups (1.7,0.85,0.42 g.kg-1), model group (0.2 mL pure water) and Guanxin Danshen capsule group (0.35 g.kg- 1), after continuous ig administration for 14 days, the trichloromethane mouse arrhythmia model was duplicated to observe the incidence of ventricular fibrillation in mice. 50 rats were randomly divided into 5 groups: Shuxin Tang high, medium and low dose groups (1.18,0.59,0.29 g.kg-1), model group (2 mL pure water) and Guanxin Danshen capsule group (0.24 g.kg -1) for ig administration for 14 days, the model of arrhythmia caused by aconitine was established and the protective effect of different doses of Shuxin Decoction on arrhythmia of model animals was observed. 60 rats were randomly divided into Shuxin Tang high, medium and low dose groups, blank control group, coronary heart Danshen capsule group and model group, the dose, the method above, the blank control group were given the same model group, continuous administration 14 d, 12 d began to use isoproterenol to establish rat model of myocardial ischemia, continuous modeling 2 d, observe the protective effect of Xinshu decoction on myocardial ischemia. Results: The high-dose Xin-shu decoction decreased the incidence of ventricular fibrillation (VF) induced by chloroform in mice to 25%; increased ventricular premature beat (VP) and ventricular tachycardia (VT ), The dosage of aconitine (VF) was 19.7,23.7,32.6μg.kg-1, the amplitude of T wave in ECG was decreased, and the levels of creatine kinase (CK) and aspartate aminotransferase AST) content reduced to 2 572.9,111.3 UL-1. Conclusion: Xinshu decoction has protective effect on heart rhythm abnormality and myocardial ischemia.
