目的了解CD147与胆管癌侵袭转移的关系。方法通过RT-PCR和免疫荧光法了解CD147在胆管癌细胞株QBC939中的表达,然后构建反义CD147分子基因片断的重组腺病毒抑制胆管癌细胞株CD147分子的表达,观察其基质金属酶MMPs改变及生长情况。结果 CD147分子在胆管癌细胞株QBC939中表达,表明反义CD147通过抑制肿瘤细胞的CD147分子的表达,主要减少了肿瘤组织中基质金属酶MMP-2,而不是MMP-9的表达,导致肿瘤的生长抑制。结论 CD147分子的高表达能够促进胆管癌的肿瘤生长,并且CD147促进肿瘤生长的能力可能与MMP-2的高表达密切相关。
Objective To understand the relationship between CD147 and invasion and metastasis of cholangiocarcinoma. Methods The expression of CD147 in cholangiocarcinoma cell line QBC939 was detected by RT-PCR and immunofluorescence. The recombinant adenovirus expressing antisense CD147 gene fragment was used to inhibit the expression of CD147 in cholangiocarcinoma cell line and the changes of MMPs were observed. And growth conditions. RESULTS: The CD147 molecule was expressed in the cholangiocarcinoma cell line QBC939, indicating that antisense CD147 mainly reduced the expression of MMP-2, but not MMP-9, in the tumor tissue by inhibiting the expression of the CD147 molecule in tumor cells, resulting in tumor Growth inhibition. Conclusion The high expression of CD147 can promote the tumor growth of cholangiocarcinoma, and the ability of CD147 to promote tumor growth may be closely related to the high expression of MMP-2.
