[目的]探讨RhoA蛋白在胃癌细胞核中的表达及预后意义。[方法]选取有详细临床资料的胃癌组织179例及正常胃黏膜组织80例,应用免疫组织化学改良二步法检测RhoA蛋白的胞核表达。[结果]RhoA蛋白在胃癌及正常胃黏膜组织细胞核中的表达阳性率分别为69.8%(125/179)和25.0%(20/80),差异有显著性(P<0.05)。RhoA蛋白胞核表达率与胃癌肿瘤分化、TNM分期、淋巴结转移有关(P<0.05)。RhoA胞核阳性患者的术后生存期短于阴性患者(P=0.022)。Cox风险比例模型表明,RhoA胞核阳性是胃癌术后预后的独立指标(OR=3.09,P=0.005)。[结论]RhoA蛋白在胃癌细胞核中过表达,是判断胃癌患者术后生存期的独立预后指标。
[Objective] To explore the expression of RhoA protein in gastric cancer cell nuclei and its prognostic significance. [Methods] 179 cases of gastric cancer tissues and 80 cases of normal gastric mucosa tissues with detailed clinical data were selected and the nucleus expression of RhoA protein was detected by an improved immunohistochemical two-step method. [Results] The positive rates of RhoA protein expression in gastric nucleus and normal gastric mucosa were 69.8% (125/179) and 25.0% (20/80), respectively, and the difference was significant (P<0.05). The nuclear expression rate of RhoA protein was related to tumor differentiation, TNM staging and lymph node metastasis of gastric cancer (P<0.05). The postoperative survival of RhoA nuclear positive patients was shorter than that of negative patients (P=0.022). Cox proportional hazards model showed that nuclear positive RhoA was an independent indicator of prognosis of gastric cancer (OR=3.09, P=0.005). [Conclusion] Overexpression of RhoA protein in gastric cancer cell nuclei is an independent prognostic indicator for judging the postoperative survival of gastric cancer patients.
