目的探讨青蒿素体外抗柯萨奇B组3型病毒(CVB3)的作用。方法以CVB3感染HeLa细胞为实验模型,采用微量细胞病变抑制法、半定量RT鄄PCR实验和间接免疫荧光实验的方法,观察分析青蒿素对CVB3的直接灭活、阻断吸附和抑制复制的作用,以及青蒿素对CVB3在感染细胞内核酸复制和蛋白表达的影响。结果青蒿素不同程度地阻断病毒吸附和抑制病毒复制,明显抑制CVB3核酸复制与蛋白表达,未发现有直接灭活病毒的作用。结论青蒿素类药物具有体外抗CVB3的作用,其抗病毒机制是通过阻断病毒吸附和抑制病毒复制来完成的。
Objective To investigate the effect of artemisinin on coxsackie B virus type 3 (CVB3) in vitro. Methods HeLa cells infected by CVB3 were used as the experimental model. The effects of artemisinin on the direct inactivation of CVB3, blockade of adsorption and inhibition of replication were observed using the methods of cytopathic inhibition, semi-quantitative RT PCR and indirect immunofluorescence. Effects, and the effect of artemisinin on the nucleic acid replication and protein expression of CVB3 in infected cells. RESULTS Artemisinin blocked the adsorption of virus and inhibited virus replication in different degrees, and significantly inhibited CVB3 nucleic acid replication and protein expression. No direct inactivation of virus was found. Conclusion Artemisinin drugs have anti-CVB3 activity in vitro, and its antiviral mechanism is accomplished by blocking virus adsorption and inhibiting virus replication.
