目的:考察山楂中熊果酸的提取纯化方法,为含有山楂的临床应用剂型研究提供科学数据。方法:首先采用L_9(3~4)的正交试验设计,确定了山楂中熊果酸为主要有效成分的提取最佳工艺;采用酸碱沉淀法富集,用X-5大孔树脂纯化,通过梯度洗脱过程中洗脱剂组合的优化进行最佳洗脱。结果:最佳提取工艺为用95%的乙醇回流提取3次,每次加8倍量、提取2h,其中回流提取次数是主要影响因素,提取率达92.9%;确定了最佳的洗脱剂组合为去离子水-30%乙醇-50%乙醇-70%乙醇-90%乙醇(pH=11)。结论:富集方法可行,熊果酸纯度可达52.3%。
Objective: To investigate the method of extraction and purification of ursolic acid in hawthorn and provide scientific data for the clinical application of the formulation containing hawthorn. Methods: Firstly, orthogonal design of L_9 (3-4) was used to determine the optimal extraction process of ursolic acid from hawthorn as the main active ingredient. The product was acid-base precipitation and purified with X-5 macroporous resin. Optimum elution was achieved by optimizing the combination of eluents during the gradient elution. Results: The optimum extraction process was extraction with 95% ethanol for 3 times, 8 times for each extraction and extraction for 2 hours. The number of reflux extraction was the main influencing factor, the extraction rate was 92.9%. The best eluent The combination is deionized water - 30% ethanol - 50% ethanol - 70% ethanol - 90% ethanol (pH = 11). Conclusion: Enrichment method is feasible, ursolic acid purity up to 52.3%.
