把抗癌药物注入蛛网膜下腔以治疗脑部恶性肿瘤,以前曾试用过,但从治疗效果及安全性方面探讨注药后药动学,尤其是药物对正常脑组织渗透性尚无报告。作者采用此给药法,并对其安全性进行了动物试验,选择对正常脑组织无毒性,又可取得一定疗效的剂量,并用自动摄影法对ACNU 注药后在脑内的分布进行了研究。方法体重150g 的正常wistar 种大白鼠,在全麻下经皮穿刺向小脑延髓池内注入以0.1ml 蒸馏水溶解的ethylene~(-14)C—ACNU1μCi,然后于5分、30分及3小时杀死。每组3只,立即取出脑放入弗立昂(Freon)内速冻,在冷却到-18℃的低温恒温器内制成厚20μm
