红霉素栓的效价测定方法,在用无水乙醇研磨稀释时,因其基质大量析出或乳化,使试验结果参差不齐,经改变溶剂量及过滤,多次试验仍不能克服上述存在的问题。经摸索,现改用氯仿作溶剂,待样品在水浴上融化后直接加氯仿,再用无水乙醇稀释的方法。经多次试验,不仅方法较快速简便,而且结果重现性也好。 一、改进后的效价测定方法: 取本品5粒(20万μ/粒或10万μ/粒),放入100ml容量瓶中,在40℃水浴中熔化后,加入氯仿约60ml,摇匀,放置至室温,再用氯仿稀释至刻度。精密吸取上述氯仿液2ml,置100ml容量瓶中,用无水乙醇稀释至刻度。照红霉素项下(中国药典1977年版)测定即得。结果见下表:
