目的探讨丹参酮ⅡA对糖尿病大鼠视网膜细胞氧化损伤及视网膜细胞凋亡的影响。方法选取5~7周健康雄性Wistar大鼠65只,随机选择50只一次性向大鼠腹腔内注射液链脲佐菌素60mg/kg诱导大鼠糖尿病,选取其中其中44糖尿病大鼠随机分为糖尿病组和给药组。选择同批次同种属大鼠15只作为对照组。给药组给予丹参酮ⅡA30mg/kg灌胃,每天1次;糖尿病组和对照组给予同体积的生理盐水灌胃,每天1次。三组大鼠均连续给药或生理盐水90天。喂养90天后测定三组大鼠的血糖水平、血中自由基含量、视网膜组织中MDA含量和超氧化物歧化酶活性、视网膜组织细胞凋亡情况。结果糖尿病组血糖水平、SOD活性高于对照组,差异有统计学意义(P<0.05);糖尿病组全血中自由基含量、MDA含量低于对照组,差异有统计学意义(P<0.05)。糖尿病组大鼠视网膜细胞溶解液A值高于对照组,差异有统计学意义(P<0.05);给药组大鼠视网膜细胞溶解液A值低于对照组,差异有统计学意义(P<0.05)。结论丹参酮ⅡA对糖尿病大鼠视网膜损伤有一定保护作用,可能与其抗氧化损伤及抑制视网膜组织细胞凋亡有关。
Objective To investigate the effects of Tanshinone ⅡA on oxidative damage of retinal cells and apoptosis of retinal cells in diabetic rats. Methods Sixty-five healthy male Wistar rats were selected from 5 to 7 weeks. Fifty rats were randomly selected to induce streptozotocin 60 mg / kg intraperitoneal injection in rats. 44 diabetic rats were randomly divided into diabetic Group and administration group. Select the same batch of 15 rats of the same species as a control group. TanshinoneⅡA30mg / kg was given to the treatment group by gavage once a day, while the diabetic group and the control group were given the same volume of normal saline once a day. Three groups of rats were given continuous or saline for 90 days. After 90 days of feeding, blood glucose level, free radical content in blood, MDA content in the retina tissue, superoxide dismutase activity and retinal cell apoptosis were measured. Results The level of blood glucose and SOD in diabetic group were significantly higher than those in control group (P <0.05). The contents of free radicals and MDA in whole blood of diabetic group were lower than those in control group (P <0.05) . The A value of retinal cell lysate in diabetic rats was higher than that in control group, the difference was statistically significant (P <0.05). The A value of retinal cell lysate in diabetic rats was lower than that in control group (P < 0.05). Conclusion Tanshinone ⅡA can protect retinal damage in diabetic rats, which may be related to its anti-oxidative damage and inhibition of retinal cell apoptosis.
