实现原代肝细胞的冻存可以降低实验研究对新鲜肝组织的需求,冻存的肝细胞可以随时制备、保存并方便应用。由于人肝来源的限制,冻存人肝细胞显得特别重要。成功冻存肝细胞的关键包括缓慢加入冻存液、控制冻存的温度、温度下降的速率、防止冰晶的形成、-150℃储存及快速解冻等。研究表明冻存的肝细胞在短期异物代谢和毒性评价中是有应用价值的。本文对肝细胞冻存和解冻过程以及冻存肝细胞的应用进行了综述。
Realizing the cryopreservation of primary hepatocytes can reduce the need for experimental studies on fresh liver tissue. Frozen hepatocytes can be prepared, preserved and conveniently applied. Cryopreserved human hepatocytes are of particular importance due to the limitations of human liver origin. Key to successful cryopreservation of hepatocytes include slow addition of cryopreservation fluid, control of cryopreservation temperature, rate of temperature drop, prevention of ice crystal formation, storage at -150 ° C and rapid thawing. Studies have shown that frozen hepatocytes are valuable in the assessment of short-term foreign body metabolism and toxicity. This review summarizes the process of freezing and thawing of hepatocytes and the application of cryopreserved hepatocytes.
