目的:探讨降糖消渴颗粒对高脂饮食诱导2型糖尿病KKAy小鼠糖脂代谢的影响。方法:30只KKAy小鼠予以高脂饲料喂养4周后,建立2型糖尿病(空腹血糖≥13.9 mmol·L~(-1))小鼠模型。将成模后的KKAy小鼠随机分为模型组、吡格列酮组、降糖消渴颗颗粒高、中、低剂量组(按生药量计为1.75、3.5、7 g·kg~(-1)),6只C57BL/6J鼠为正常对照组。干预10周后,称取体质量和肝重,计算肝重指数;检测小鼠血清和肝脏中甘油三酯、胆固醇、高密度脂蛋白、低密度脂蛋白的含量。结果:降糖消渴颗粒低剂量(1.75 g·kg~(-1))可显著降低血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL)(P<0.01),且可降低肝脏中甘油三酯(TG)含量(P<0.05);中剂量(3.5g·kg~(-1))显著降低了糖尿病小鼠的肝重指数,对血清和肝脏脂质各项指标都具有良好的改善作用(P<0.01);高剂量(7 g·kg~(-1))不仅降低了糖尿病小鼠的肝重指数(P<0.01),对血清中TG、LDL及肝脏脂质各项指标也有显著改善。结论:降糖消渴颗粒可以有效改善T2DM状态下肝脏的脂质代谢紊乱,降低血脂含量。
Objective: To investigate the effects of Jiang Tang Xiaoke Granule on glucose and lipid metabolism in type 2 diabetic KKAy mice induced by high-fat diet. Methods: Thirty KKAy mice were fed with high-fat diet for 4 weeks, and mice model of type 2 diabetes (fasting blood glucose≥13.9 mmol·L -1) were established. The KKAy mice were randomly divided into model group, pioglitazone group, high, medium and low dosage of Jiangtang Xiaoke Granule (1.75, 3.5, 7 g · kg -1) Six C57BL / 6J mice were normal control group. After intervention for 10 weeks, the body weight and liver weight were weighed and the liver weight index was calculated. The content of triglyceride, cholesterol, high density lipoprotein and low density lipoprotein in serum and liver of mice were measured. Results: The low dose (1.75 g · kg -1) of Jiangtang Xiaoke Granule could significantly reduce the serum total cholesterol (TC) and low density lipoprotein cholesterol (LDL) (P <0.01), and decrease the content of glycerol The triglyceride (TG) content (P <0.05) and the middle dose (3.5g · kg -1) significantly reduced the liver index of diabetic mice, and had a good improvement on various indexes of serum and liver lipid (P <0.01). High dose (7 g · kg -1) not only reduced the liver weight index (P <0.01) in diabetic mice, but also had significant correlation with the serum TG, LDL and liver lipid Significantly improved. Conclusion: Jiangtangxieke granule can effectively improve the liver dyslipidemia and lower the blood lipid level in T2DM.
