目的:研究甜瓜叶片Hsp70基因在白粉病胁迫下的表达差异。方法:采用对白粉病抗性不同的2个甜瓜品种,Trizol法提取叶片总RNA,PCR技术克隆Hsp70基因核心片段,采用Semi-Q-RT-PCR技术,以Actin基因为内参基因,对白粉病胁迫下不同处理、不同品种的甜瓜叶片Hsp70基因的表达进行差异分析。结果:抗病品种MR-1与感病品种伽师瓜叶片中的Hsp70基因表达量均随着接种时间的延长逐渐减少,但MR-1接种白粉病1d后表达量稍有增加;且伽师瓜中Hsp70基因的表达差异较MR-1明显。结论:两种甜瓜叶片感染白粉病后,Hsp70表达量均减少。
Objective: To study the difference of expression of Hsp70 gene in melon leaves under powdery mildew stress. Methods: Two kinds of melon cultivars with different resistance to powdery mildew were used. The total RNA of leaves was extracted by Trizol method. The core fragment of Hsp70 gene was cloned by PCR. Semi-Q-RT-PCR technique and Actin gene were used as internal reference genes for powdery mildew Under different stress, the expression of Hsp70 in different varieties of melon leaves was analyzed by difference. Results: The expression levels of Hsp70 in resistant varieties MR-1 and susceptible cultivar Jiashi melon decreased gradually with the inoculation time, but the expression level of MR-1 slightly increased after 1d inoculation with powdery mildew; The difference of Hsp70 gene expression in melon is more obvious than MR-1. Conclusion: The expression of Hsp70 in two melon leaves infected with powdery mildew decreased.
