目的:检测新扶正除疫颗粒对肺炎链球菌所致小鼠急性上呼吸道感染模型肺组织中TLR2、TLR4mRNA变化的影响,探究新扶正除疫颗粒在抗炎及免疫方面的作用机制。方法:小鼠分为空白组、模型组、新扶正除疫颗粒的高中低剂量组、中药对照组和西药对照组,造模1h后灌药,并于造模后第1、4、7天取材,采用Real-time PCR方法测定小鼠肺组织中TLR2、TLR4 mRNA。结果:小鼠肺组织中TLR2在第1、4天各药物干预组与空白组和模型组比较,差异显著(P<0.05),第7天各组与模型组差异显著,但除低剂量组外均与空白组无显著性差异。肺组织中TLR4在第1、4天时,高、中剂量组、中药对照组及西药对照组与模型组有差异,但与空白组无显著差异。各组均存在随给药时间TLR2、TLR4 mRNA含量出现下降的趋势,新扶正除疫颗粒高、中剂量组优于低剂量组,与西药对照组无显著性差异。结论:提示新扶正除疫颗粒可以激活TLR2/TLR4系统,从而达到抗炎与免疫调节的作用,增强免疫与抗炎抑菌作用和中医“扶正”与“祛邪”的治法有关联性。
Objective: To investigate the effect of Xinfuzhengzhiqi granule on the changes of TLR2 and TLR4mRNA in the lung tissue of mice with acute upper respiratory tract infection induced by Streptococcus pneumoniae, and to explore the mechanism of action of Xifuzhengzhiqi granule in anti-inflammatory and immunological. Methods: The mice were divided into blank group, model group, high and low dose Xinfuzheng tablets, traditional Chinese medicine control group and western medicine control group, The materials were collected and the TLR2 and TLR4 mRNA in the lung of mice were detected by Real-time PCR. Results: Compared with the blank group and the model group, the TLR2 in the lung tissue of the mice in the first and the fourth day was significantly different (P <0.05). On the 7th day, there was significant difference between each group and the model group, except the low dose group There was no significant difference between the control group and the blank group. TLR4 in lung tissue at day 1, 4, high, medium dose group, traditional Chinese medicine control group and western medicine control group and the model group differences, but no significant difference with the blank group. In each group, TLR2 and TLR4 mRNA levels tended to decrease with the time of administration. The high and mid-dose granules of Xinfuzheng were better than the low-dose group, which showed no significant difference compared with western medicine control group. Conclusion: It is suggested that Xifuzhengqi granule can activate TLR2 / TLR4 system to achieve anti-inflammatory and immunomodulatory effects, enhance immunity, anti-inflammatory and anti-bacterial effects and cure the traditional Chinese medicine “Fuzheng” and “Qu Xie” Relevant.
