目的 观察瘦素对大鼠肝细胞蛋白激酶 C(PKC)蛋白表达的影响。方法 分离、培养新生Sprague- Dawley大鼠肝细胞 ,与瘦素 (5 nmol/ L)孵育 12 ,2 4,36 h,提取蛋白后用 Western印迹法检测各时间点蛋白激酶 C的蛋白表达水平。结果 大鼠肝细胞 c PKCα的蛋白水平在瘦素孵育 12 h及 2 4h后同对照组相比均未发生显著变化 (P>0 .0 5 ) ;但在孵育 36 h后同对照组相比显著升高 (P<0 .0 5 )。经瘦素孵育 2 4及 36 h后 ,大鼠肝细胞 c PKCα的蛋白水平同孵育 12 h时相比亦显著升高 (P<0 .0 5 )。结论 瘦素可上调大鼠肝细胞蛋白激酶 C的蛋白表达 ,这种促进作用呈时间依赖性。
Objective To observe the effect of leptin on protein kinase C (PKC) protein expression in rat liver cells. Methods Hepatic cells of neonatal Sprague-Dawley rats were isolated and incubated with leptin (5 nmol / L) for 12,24 and 36 h. The protein expression levels of protein kinase C at each time point were detected by Western blotting. Results The protein level of c PKCα in rat hepatocytes did not change significantly at 12 h and 24 h after leptin treatment (P> 0.05), but compared with the control group at 36 h after incubation Significantly increased (P <0.05). After 24 and 36 h of leptin incubation, the protein level of c PKCα in rat hepatocytes increased significantly compared with that at 12 h after incubation (P <0.05). Conclusion Leptin can upregulate protein kinase C protein expression in rat liver cells in a time-dependent manner.
