为了探讨柠檬酸对机体的毒副作用,选用昆明系健康雄性小鼠40只,随机分成4组,每组10只,饲养1周后,腹腔注射一定剂量的生理盐水及低(180 mg.kg-1)、中(270 mg.kg-1)、高(360 mg.kg-1)质量浓度的柠檬酸溶液.每周注射1次,连续注射3周,第3次注射后的第7天,所有小鼠均颈椎脱臼处死,采取小鼠的脑组织分别于液氮和-20℃各保存1份备用.通过RT-PCR法和ELISA法分别检测小鼠脑组织中bcl-2基因表达水平和蛋白表达量.结果显示,柠檬酸处理组与对照组相比,bcl-2基因表达水平均无统计学上的差异(P>0.05);中剂量柠檬酸处理时,bcl-2蛋白的表达量达到最高值,且与对照组呈现显著差异(P<0.05),上升了23%,柠檬酸高剂量组与中剂量组相比,bcl-2蛋白的表达量呈显著降低(P<0.05);其它组间均无统计学上的差异(P>0.05).说明腹腔注射270 mg.kg-1的柠檬酸可显著提高小鼠脑组织bcl-2蛋白的表达量;但本试验设置的柠檬酸梯度对bcl-2基因表达水平无显著影响.
In order to investigate the toxic and side effects of citric acid on the body, 40 male Kunming healthy male mice were randomly divided into 4 groups with 10 rats in each group. The expression of bcl-2 protein in high dose citric acid group was significantly lower than that in middle dose group (P <0.05). (P> 0.05), indicating that intraperitoneal injection of citric acid at a dose of 270 mg.kg-1 significantly increased the expression of bcl-2 protein in mouse brain tissue. However, in this experiment, Citrate gradient no significant effect on bcl-2 gene expression.
