目的研究壁虎多肽混合物(GPM)对人食管癌EC109细胞的生长抑制作用及其机制。方法用MTT法检测GPM对细胞增殖的影响;Hoechst 33258荧光染色法观察GPM对细胞形态的影响;免疫细胞化学技术检测细胞内Bcl-2、Caspase-3的表达。结果 GPM抑制了EC109细胞的生长,作用呈时间和剂量依赖性。GPM作用24 h,EC109细胞出现明显的凋亡,且Bcl-2表达降低而Caspase-3表达增高。结论 GPM可以抑制人食管癌EC109细胞增殖并诱导其凋亡,其机制可能与GPM降低食管癌EC109细胞Bcl-2表达而增加Caspase-3表达有关。
Objective To study the inhibitory effect of gecko peptide mixture (GPM) on human esophageal carcinoma EC109 cells and its mechanism. Methods MTT assay was used to detect the effect of GPM on cell proliferation. Hoechst 33258 staining was used to observe the effect of GPM on cell morphology. Immunocytochemistry was used to detect the expression of Bcl-2 and Caspase-3. Results GPM inhibited the growth of EC109 cells in a time and dose-dependent manner. At 24 h after GPM treatment, EC109 cells showed obvious apoptosis, and the expression of Bcl-2 and Caspase-3 increased. Conclusion GPM can inhibit the proliferation and induce the apoptosis of human esophageal carcinoma EC109 cells, which may be related to the decrease of Bcl-2 expression and the increase of Caspase-3 expression induced by GPM in esophageal carcinoma EC109 cells.
