为观察卡介菌多糖核酸 (BCG_PSN)对哮喘患儿外周血单个核细胞 (PBMC)Th1/Th2细胞因子分泌的调节作用 ,并与地塞米松作比较。对25例哮喘患儿 ,取外周血分离单个核细胞。其中21例患儿的PBMC同时分成对照组和BCG_PSN组 ;19例患儿的PBMC同时分成对照组和地塞米松组 ,培养48小时。ELISA法测定培养上清液中的γ干扰素 (IFN_γ)、白细胞介素4(IL_4)的水平。结果显示 ,BCG_PSN可明显增高IFN_γ的分泌水平 ,地塞米松则抑制之 ;BCG_PSN对IL_4的分泌无明显影响 ,而地塞米松明显抑制IL_4分泌 ;BCG_PSN通过增高Th1型细胞因子 (IFN_r) ,而地塞米松通过抑制Th2型细胞因子(IL_4)的分泌 ,两者均提高Th1/Th2(IFN_r/IL_4)比值。提示BCG_PSN可使Th1/Th2平衡向Th1型转化 ,调节免疫失衡 ,从而起到治疗哮喘的作用
To investigate the regulatory effect of BCG_PSN on the secretion of Th1 / Th2 cytokines in peripheral blood mononuclear cells (PBMCs) from asthmatic children and to compare with dexamethasone. Peripheral blood mononuclear cells were isolated from 25 asthmatic children. PBMCs of 21 children were divided into control group and BCG_PSN group at the same time. PBMC of 19 children were divided into control group and dexamethasone group for 48 hours. The levels of IFN-γ and IL-4 in culture supernatants were determined by ELISA. The results showed that BCG_PSN significantly increased the secretion of IFN_γ and dexamethasone inhibited it; BCG_PSN had no significant effect on the secretion of IL_4, while dexamethasone significantly inhibited the secretion of IL_4; BCG_PSN increased Th1-type cytokines (IFN_r) Both of them increased Th1 / Th2 (IFN_r / IL_4) ratio by inhibiting the secretion of Th2 cytokines (IL_4). Tip BCG_PSN can Th1 / Th2 balance to Th1-type conversion, regulating immune imbalance, which play a role in the treatment of asthma
