目的:正交优化甘草中甘草酸的提取工艺。方法:采用紫外分光光度法建立甘草酸含量的测量方法;考察提取时间、提取次数、提取温度、液固比和乙醇浓度对提取效果的影响;在单因素实验的基础上进行正交试验,采用正交表L9(34)优化甘草酸的提取工艺。结果:建立甘草酸的含量测定方法;通过考察5个单因素,结合正交试验优化得到甘草酸的最佳提取工艺条件为:提取温度为50℃,液固比为8∶1,提取次数为3次(每次为1h)。结论:本实验优化的甘草酸提取工艺简单稳定,成本低,适用于工业化生产,可为含甘草酸的制剂工艺研究提供参考。
Objective: To optimize the extraction of glycyrrhizic acid from licorice by orthogonal design. Methods: The method of determination of glycyrrhizic acid content was established by ultraviolet spectrophotometry. The effects of extraction time, extraction times, extraction temperature, liquid-solid ratio and ethanol concentration on the extraction efficiency were studied. The orthogonal experiment was carried out on the basis of single factor experiment. Orthogonal Table L9 (34) Optimization of glycyrrhizic acid extraction process. Results: The determination method of glycyrrhizic acid was established. The optimal extraction conditions for glycyrrhizic acid were as follows: the extraction temperature was 50 ℃, the liquid-solid ratio was 8:1, the number of extraction was 3 times (each time for 1h). Conclusion: The glycyrrhizic acid extraction process optimized in this experiment is simple and stable, low cost, suitable for industrial production, which can provide reference for the preparation of glycyrrhizin-containing preparations.
