目的探索弥漫性大B细胞淋巴瘤(DLBCL)BALB/C-nu/nu裸鼠皮下移植瘤模型建立的方法。方法将DLBCL细胞株SUDHL-10、OCI-Ly8、OCI-Ly10接种于BALB/C-nu/nu裸鼠腋部皮下,每2~3天观察皮下肿瘤体积变化,并绘制肿瘤生长曲线。在裸鼠濒临死亡或皮下肿瘤长径≥20mm时将裸鼠处死,分离肿瘤组织进行HE染色以及免疫组化染色。结果裸鼠皮下见有肿瘤长出的时间为接种后第10~12天;SUDHL-10、OCI-Ly8、OCI-Ly10细胞接种的成瘤率分别为66.7%(4/6)、100%(6/6)、83.3%(5/6);接种后第42天皮下肿瘤长径最大可达20mm,此时裸鼠逐渐出现濒死状态,SUDHL-10、OCI-Ly8、OCI-Ly10细胞接种的肿瘤体积分别为(2617.66±332.98)mm~3、(2679.66±367.02)mm~3、(2409.60±640.84)mm~3。移植瘤HE染色形态与人DLBCL相符合。免疫组化染色显示,SUDHL-10、OCI-Ly8细胞接种后的肿瘤细胞CD20弥漫阳性、CD10弥漫阳性、Bcl-6阳性;OCI-Ly10细胞接种后的肿瘤细胞CD20部分阳性、CD10弥漫阳性、Bcl-6阴性、多发性骨髓瘤癌基因1阴性。结论成功构建DLBCL细胞BALB/C-nu/nu裸鼠皮下移植瘤模型;其移植瘤细胞与人DLBCL细胞相似,是较理想的研究DLBCL的实验动物模型。
Objective To explore a method for establishing a subcutaneous xenograft model of BALB / C-nu / nu nude mice with diffuse large B cell lymphoma (DLBCL). Methods DLBCL cell lines SUDHL-10, OCI-Ly8 and OCI-Ly10 were inoculated subcutaneously in the axilla of BALB / C nu nu mice. The volume of subcutaneous tumors was observed every 2 to 3 days and the tumor growth curve was drawn. The nude mice were sacrificed on the verge of death or when the subcutaneous tumor diameter ≥20 mm. The tumor tissues were separated for HE staining and immunohistochemical staining. Results Tumor growth in nude mice was observed on the 10th to 12th day after inoculation. The tumorigenic rates of SUDHL-10, OCI-Ly8 and OCI-Ly10 cells were 66.7% (4/6) and 100% 6) and 83.3% (5/6), respectively. On the 42nd day after inoculation, the maximum diameter of the subcutaneous tumors was up to 20mm. At this time, the nude mice gradually became moribund. SUDHL-10, OCI-Ly8 and OCI-Ly10 cells were inoculated The tumor volume was (2617.66 ± 332.98) mm ~ 3, (2679.66 ± 367.02) mm ~ 3 and (2409.60 ± 640.84) mm ~ 3, respectively. The morphology of xenografts was consistent with human DLBCL. Immunohistochemical staining showed that CD20 was diffusely positive in tumor cells after inoculation of SUDHL-10 and OCI-Ly8 cells, but diffusely positive for CD10 and positive for Bcl-6. The CD20 of tumor cells after OCI-Ly10 cells inoculation was partially positive, CD10 diffusely positive, Bcl- -6 negative, multiple myeloma oncogene 1 negative. Conclusion The subcutaneous xenograft models of BALB / C-nu / nu mice with nude mice bearing DLBCL were successfully constructed. The transplanted tumor cells resembled human DLBCL cells, which is an ideal experimental animal model for DLBCL.
