目的研究白藜芦醇对小鼠慢性哮喘的作用。方法小鼠鼻内滴注螨虫(HDM)粗提物25μg,每周5次,连续7周,制备小鼠过敏性哮喘模型。将哮喘小鼠随机分为实验组(白藜芦醇100 mg·kg~(-1)),对照组(地塞米松1 mg·kg~(-1)),另设正常组及模型组(均0.9%Na Cl),每组15只。2周后,称量小鼠体重,以酶联免疫吸附法测定小鼠肺灌流液(BALF)中细胞因子含量;观察小鼠肺组织的病理学变化。结果在小鼠的BALF中,模型组的白细胞介素-17(IL-17)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、肿瘤生长因子-β(TGF-β)分别为(186.0±101.0)pg·mL~(-1),(29.6±10.6)ng·mL~(-1),(11.1±3.2)ng·mL~(-1),(158.5±10.0)ng·mL~(-1)。实验组的IL-17、IL-6、TNF-α、TGF-β分别为(43.1±10.3)pg·mL~(-1),(10.4±1.4)ng·mL~(-1),(5.0±1.7)ng·mL~(-1),(107.8±17.3)ng·mL~(-1)。与模型组相比,白藜芦醇能明显抑制BALF中IL-17、IL-6、TNF-α、TGF-β的表达,差异有统计学意义(P<0.05)。实验组对小鼠体重的改变没有影响。结论白藜芦醇对螨虫诱导的小鼠慢性哮喘具有一定的改善作用。
Objective To study the effect of resveratrol on chronic asthma in mice. Methods Mice were intranasally instilled with 25 μg of mite (HDM) crude extract for 5 weeks for 7 consecutive weeks to prepare mouse allergic asthma model. The asthmatic mice were randomly divided into experimental group (resveratrol 100 mg · kg -1) and control group (dexamethasone 1 mg · kg -1), normal group and model group 0.9% NaCl), 15 in each group. After 2 weeks, the body weight of the mice was weighed and the content of cytokines in the lung perfusion fluid (BALF) of the mice was measured by enzyme-linked immunosorbent assay. The pathological changes in the lung tissues were observed. Results In the BALF of mice, the levels of IL-17, IL-6, TNF-α and tumor growth factor-β TGF-β were (186.0 ± 101.0) pg · mL -1, 29.6 ± 10.6 ng · mL -1, 11.1 ± 3.2 ng · mL -1, 158.5 ± 10.0 ng · mL -1. The levels of IL-17, IL-6, TNF-α and TGF-β in the experimental group were (43.1 ± 10.3) pg · mL -1, 10.4 ± 1.4 ng · mL -1, ± 1.7 ng · mL -1 and 107.8 ± 17.3 ng · mL -1, respectively. Compared with model group, resveratrol could significantly inhibit the expression of IL-17, IL-6, TNF-α and TGF-β in BALF with statistical significance (P <0.05). The experimental group had no effect on the weight change of the mice. Conclusion Resveratrol can improve the chronic asthma induced by mites in mice.
