目的探讨阿魏酸钠(Sodium ferulate,SF)对大鼠纤维化心肌组织中结缔组织生长因子(Connective tissue growth factor,CTGF)表达的影响。方法一次性皮下注射盐酸异丙肾上腺素(Isoproterenol,Iso)15mg/kg体重,复制Wistar大鼠心肌缺血性坏死模型,于不同时间点脱臼处死大鼠,取心脏,采用半定量RT-PCR法检测大鼠组织中CTGF基因mRNA的转录水平,免疫组化法检测CTGF蛋白的表达水平。再次复制大鼠心肌缺血坏死模型,同时注射SF进行同步干预,3周后检测大鼠心肌组织中CTGF基因mRNA的转录水平及蛋白的表达水平。结果注射Iso后24h,CTGF基因mRNA的转录水平达高峰,较正常对照组明显增加,3周时仍高于正常对照组;CTGF蛋白的表达随纤维化病变程度的加重而增加。SF干预后,CTGF基因mRNA的转录水平及蛋白的表达水平均明显下降。结论 CTGF的表达与大鼠心肌纤维化(Myocardial fibrosis,MF)程度密切相关,提示CTGF可能在MF中起重要作用;SF对CTGF的表达具有抑制作用。
Objective To investigate the effect of sodium ferulate (SF) on the expression of connective tissue growth factor (CTGF) in rat cardiac fibrosis. Methods Fifteen mg / kg body weight of Isoproterenol (Iso) was subcutaneously injected subcutaneously into the Wistar rat model of myocardial ischemic necrosis. The rats were sacrificed at different time points, and the heart was taken. The semi-quantitative RT-PCR The mRNA level of CTGF mRNA in rat tissues was detected, and the expression of CTGF protein was detected by immunohistochemistry. The model of myocardial ischemia and necrosis was reproduced again. At the same time, SF was injected simultaneously for simultaneous intervention. The mRNA and protein expression of CTGF mRNA in myocardium were detected 3 weeks later. Results The mRNA level of CTGF reached a peak at 24 hours after injection of Iso, which was significantly higher than that of the normal control group and higher than that of the normal control group at 3 weeks. The expression of CTGF protein increased with the severity of fibrosis. SF intervention, CTGF mRNA expression and protein expression levels were significantly decreased. Conclusions The expression of CTGF is closely related to the degree of Myocardial Fibrosis (MF) in rats, suggesting that CTGF may play an important role in MF. SF may have an inhibitory effect on the expression of CTGF.
