目的 以hTERT反义核酸抑制白血病细胞(HL-60和K562)端粒酶活性,研究CDDP诱导凋亡敏感性的变化。方法 全硫代反义核酸由上海生物化学研究所合成和纯化;端粒酶活性用试剂合测定(宝灵曼公司产品);用形态学方法和流式细胞仪检测细胞调亡。结果 实验结果显示,hTERT全硫代反义核酸,通过下调hTERT基因表达,显著地抑制端粒酶活性;端粒酶活性下降以后,白血病胞对CDDP诱导调亡的敏感性显著升高。结论 以hTERT基因反义核酸抑制端粒酶活性增加白血病细胞对CDDP诱导凋亡的敏感性。
Objective To study the effect of hTERT antisense oligodeoxynucleotide on the telomerase activity of leukemia cells (HL-60 and K562) and to investigate the sensitivity of CDDP-induced apoptosis. Methods Total thio-antisense nucleic acid was synthesized and purified by Shanghai Institute of Biochemistry. The activity of telomerase was determined by Reagent kit (product of Böhrenman). Morphology and flow cytometry were used to detect apoptosis. Results The results showed that hTERT antisense oligonucleotide significantly inhibited telomerase activity by down-regulating the expression of hTERT gene. After the telomerase activity decreased, the sensitivity of leukemic cells to CDDP-induced apoptosis was significantly increased. Conclusion Inhibition of telomerase activity by hTERT antisense oligonucleotides enhances the sensitivity of leukemic cells to CDDP-induced apoptosis.
