目的分析肾移植患者 CD4+辅助性 T细胞( Th)对供者和对照者的 HLA抗原的应答状态。方法采集 16位长期( 3~ 15年)携带功能良好移植肾的患者外周血单个核细胞( PBMC),用限制性稀释法( LDA)对 PBMC中能够识别供者或对照者细胞的 Th细胞的比率( Th- p)进行分析。结果 8例患者的 PBMC对供者细胞处于明显的低反应状态,即已形成对供者 HLA特异的免疫耐受。另 8例患者则没有观察到同样的现象。但是这 8例患者 PBMC中对供者细胞特异的 Th细胞对 CD4抗体十分敏感。结论有些长期携带功能良好移植肾的患者已经获得对供者细胞特异的免疫耐受状态。没有出现供者特异的低反应状态的患者,其 T细胞受体对供者 HLA的亲合力偏低。
Objective To analyze the response of CD4 + helper T cells (Th) to HLA antigens in donor and control subjects in renal transplant recipients. Methods Peripheral blood mononuclear cells (PBMCs) from 16 patients with long-term (3-15 years) renal transplant recipients were collected. PBMCs were used to detect Th cells in PBMC from donor or control subjects by restriction endonuclease assay (LDA) Ratio (Th-p) for analysis. Results PBMCs of eight patients showed obvious low response to donor cells, that is, HLA-specific immune tolerance had been formed. The other eight patients did not observe the same phenomenon. However, the Th cells specific to donor cells in PBMC from these 8 patients were very sensitive to CD4 antibodies. Conclusions Some patients who have long-term well-functioned kidney grafts have acquired immune-specific status to donor cells. Patients without a donor-specific low-response status have low T-cell receptor affinity for donor HLA.
