目的 探讨血热型银屑病患者外周血单个核细胞组蛋白乙酰化酶(HATs)和去乙酰化酶(HDACs)的关系;研究凉血消风汤对HATs和HDACs平衡的影响,从而揭示银屑病乙酰化内稳态的变化.方法 收集寻常性血热型银屑病患者,设立中药治疗组和健康对照组,治疗组给予凉血消风汤6g,2次/d,治疗4周后,采用酶联免疫吸附法分别测定治疗前后血清中TNF-α含量,PBMC提取组蛋白后应用组蛋白乙酰化酶活性和去乙酰化酶活性试剂盒测定组蛋白HATs和HDACs水平.结果 银屑病患者治疗后血清中TNF-α表达降低(P＜0.05),HATs表达上升(P＜0.05),HDACs表达降低(P＜0.05),差异均有统计学意义.结论 凉血消风汤能够降低银屑病患者血清TNF-α含量,从而治疗银屑病,其机制可能与调节乙酰化内稳态平衡有关.“,”Objective To investigate the relationship between histone acetyltransferase(HATs) and histone deacetylase (HDACs) in the peripheral blood mononuclear cells (PBMC) of patients with psoriasis of blood heat type;study the effect of Liangxue Xiaofeng Tang on the HATs and HDACs and reveal changes in psoriasis acetylation homeostasis.Methods The research method was to set up a traditional Chinese medicine treatment group (treatment group) and a normal control group (control group).Patients with psoriasis vulgaris of blood heat type (the treatment group) received Liangxue Xiaofeng Tang 6g,2 times a day,for 4 weeks,and then measured the levels of serum TNF-α by enzyme linked immunosorbent assay(ELISA) before and after therapies to predicat the immunological effect of Liangxue Xiaofeng Tang on patients with psoriasis.Finally,we had determined the level of PBMC histone HATs and HDACs by using the HAT Activity Colorimetric Assay Kit and HDAC Activity Colorimetric Assay Kit before and after treatments to decide the effect of Liangxue Xiaofeng Tang on acetylation homeostasis of patients with psoriasis.Results The expression level of serum TNF-α decreased significantly (P ＜ 0.05),the expression level of HATs increased significantly (P ＜ 0.05) while the expression level of HDACs decreased significantly (P ＜ 0.05).Conclusion Liangxue Xiaofeng Tang could increase the activity of HATs,and reduce the activity of HDACs,the meantime maintain acetylationhomeostasis as well as decrease the levels of serum TNF-α in the peripheral blood of patients with psoriasis.
