目的:建立高效液相色谱(RP-HPLC)法同时测定一清颗粒中原小檗碱型生物碱的含量。方法:采用DIONEX C18(4.6 mm×250 mm,5μm)色谱柱;流动相为乙腈-0.05 mol·L-1磷酸二氢钾溶液(用磷酸调节pH值至3.0)(30∶70),流速1.0ml·min-1,检测波长350 nm。结果:小檗碱、黄连碱、巴马汀3种原小檗碱型生物碱在本文条件下,分离良好;标准曲线的相关系数(r)分别为1.0000、0.999 9、0.999 9;平均回收率(n=9)分别为101.2%,101.8%,101.7%,RSD分别为0.95%,0.48%,0.73%。结论:本方法准确,重复性良好,简单易行,适用于同时测定一清颗粒中3种原小檗碱型生物碱的含量。
OBJECTIVE: To establish a method for the simultaneous determination of protoberber berberine alkaloids in Yiqing granules by high performance liquid chromatography (RP-HPLC). METHODS: The mobile phase consisted of acetonitrile-0.05 mol·L-1 potassium dihydrogen phosphate (adjusted to pH 3.0 with phosphoric acid) (30:70) using a DIONEX C18 column (4.6 mm × 250 mm, 5 μm) ml · min-1, the detection wavelength was 350 nm. Results: The three berberine alkaloids of berberine, berberine and palmatine were well separated in this paper. The correlation coefficients (r) of the standard curve were 1.0000, 0.999 9 and 0.999 9, respectively. The average recoveries (n = 9) were 101.2%, 101.8% and 101.7% respectively, with RSDs of 0.95%, 0.48% and 0.73%, respectively. Conclusion: The method is accurate, reproducible and simple. It is suitable for simultaneous determination of three kinds of protoberberine alkaloids in Yiqing granule.
