目的探讨神昌注射液对谷氨酸(Glu)引起PC12细胞损伤的保护作用。方法细胞培养完成后随机分为5组;每组12份,除正常组外其余四组均用10mmol/L Glu作用于PC12细胞制成细胞损伤模型,留12份作对照,另三组给予不同剂量神昌注射液观察神昌注射液对其细胞形态、存活力、细胞凋亡率和IL-6、IL-8水平的影响。结果神昌注射液可有效改善谷氨酸诱导的PC12细胞的形态损伤,提高细胞存活力、降低细胞凋亡率及细胞因子IL-6、IL-8的水平,其中以中、高剂量更明显。结论神昌注射液对谷氨酸所致PC12细胞损伤具有保护作用,以中剂量组(19.4ul/ml)效果最优。机制可能与其能提高细胞存活力及抑制凋亡、降低IL-6、IL-8水平有关。
Objective To investigate the protective effects of Shenchang injection on the damage of PC12 cells induced by glutamic acid (Glu). Methods After the cell culture was completed, the cells were randomly divided into five groups. Twelve groups were divided into two groups. Except the normal group, all the four groups were treated with 10 mmol / L Glu on PC12 cells for 12 days. The effect of Shenchang injection on the cell morphology, viability, apoptosis rate and the levels of IL-6 and IL-8 were observed with dose Shenchang injection. Results Shenchang injection can effectively improve the morphological damage of glutamate-induced PC12 cells, improve cell viability and reduce the rate of apoptosis and cytokines IL-6, IL-8 levels, of which the medium and high doses of more obvious . Conclusion Shenchang injection has a protective effect on glutamate-induced injury of PC12 cells, and the medium dose group (19.4ul / ml) has the best effect. Mechanism may be related to its ability to increase cell viability and inhibit apoptosis, reduce IL-6, IL-8 levels.
