目的探讨Twist和HIF-1αmRNA在前列腺癌组织中表达及临床意义。方法应用原位杂交技术分别检测30例前列腺增生组织、30例前列腺上皮内瘤变组织及70例前列腺癌组织Twist、HIF-1αmRNA表达情况。结果①Twist、HIF-1αmRNA在30例前列腺增生组织、30例前列腺上皮内瘤变组织及70例前列腺癌组织中的表达分别为13.3%、46.6%、65.7%和16.7%、50.0%、71.4%,差异有统计学意义(χ2=23.15,P<0.01;χ2=25.57,P<0.01);Twist、HIF-1αmRNA在同组间表达差异无统计学意义(P>0.05)。②Twist及HIF-1αmRNA的表达均与前列腺癌的Gleason分级、临床分期、骨转移及组织坏死相关(P<0.05),而与年龄无关(P>0.05)。③Twist、HIF-1αmRNA在前列腺癌组织中过度表达,且两者之间呈正相关(r=0.34,P<0.01)。结论 Twist、HIF-1αmRNA在前列腺癌发生发展过程中起着重要作用,两者表达检测有利于更好地评估前列腺癌的生物学行为,提高对患者预后判断的准确性。
Objective To investigate the expression of Twist and HIF-1αmRNA in prostate cancer and its clinical significance. Methods The expression of Twist and HIF-1αmRNA in 30 cases of benign prostatic hyperplasia, 30 cases of prostatic intraepithelial neoplasia and 70 cases of prostatic carcinoma were detected by in situ hybridization. Results ① The expressions of Tist and HIF-1α mRNA were 13.3%, 46.6%, 65.7% and 16.7%, 50.0% and 71.4% respectively in 30 cases of benign prostatic hyperplasia, 30 cases of prostatic intraepithelial neoplasia and 70 cases of prostate cancer. The difference was statistically significant (χ2 = 23.15, P <0.01; χ2 = 25.57, P <0.01). There was no significant difference in expression of Twist and HIF-1αmRNA among the same groups (P> 0.05). ②Twist and HIF-1αmRNA expression were correlated with Gleason grade, clinical stage, bone metastasis and tissue necrosis of prostate cancer (P <0.05), but not with age (P> 0.05). ③Twist and HIF-1αmRNA were overexpressed in prostate cancer tissues, and there was a positive correlation between them (r = 0.34, P <0.01). Conclusions Twist and HIF-1αmRNA play an important role in the development of prostate cancer. The expression of Twist and HIF-1αmRNA can be used to evaluate the biological behavior of prostate cancer better and improve the accuracy of prognosis judgment.
