目的:采用HPLC-TOF/MS对人参中化学成分进行分析鉴别。方法:色谱柱Agilent Zorbax XDB-C18柱(4.6mm×250mm,5μm),乙腈与水梯度洗脱,梯度如下:1~35min,19%A;35~55min,19%~29%A;55~70min,29%A;70~110min,29%~40%A;110~150min,95%A。柱温为20℃,进样量20μl,流速1ml/min,飞行时间质谱配有APCI离子源,质量数扫描范围m/z100~1350。结果:共鉴别出人参药材中39个化学成分。结论:采用HPLC-TOF/MS方法一次性在一张图谱上表征出人参中的39个化学成分,为人参的药效物质基础及体内化学成分代谢与作用机制的深入研究奠定基础。
OBJECTIVE: To analyze and identify the chemical components of ginseng by HPLC-TOF / MS. METHODS: The column was eluted with an Agilent Zorbax XDB-C18 column (4.6 mm × 250 mm, 5 μm) with a gradient of acetonitrile and water. The gradient was as follows: 1 to 35 min, 19% A; 35 to 55 min, 19% to 29% 70 min, 29% A; 70-110 min, 29% -40% A; 110-150 min, 95% A. The column temperature was 20 ℃, the injection volume was 20μl and the flow rate was 1ml / min. The time-of-flight mass spectrometer was equipped with an APCI ion source with a mass scanning range of m / z100-1350. Results: A total of 39 chemical constituents in ginseng were identified. Conclusion: The HPLC-TOF / MS method is used to characterize 39 chemical constituents of ginseng on one map at a time, which lays the foundation for the further study on the pharmacological basis of ginseng and the in vivo chemical composition metabolism and mechanism.
