目的探讨维、汉两民族脂联素基因(adipose most abundant gene transcript1)+45位点及+349位点单核苷酸多态性与肥胖的关系。方法采用优化的特异性引物PCR(SSP-PCR)技术确定脂联素基因+45位点和+349位点的基因型。结果肥胖组与对照组中汉族脂联素基因+45位点GT基因型频率均高于维吾尔族,2组维吾尔族TT基因型频率均高于汉族,但差异无统计学意义(P>0.05);维吾尔族人群肥胖组和对照组的脂联素基因+349位点A、G等位基因频率分布差异具统计学意义(P<0.05)。结论维、汉两民族脂联素基因的+45位点可能存在差异,维吾尔族人群脂联素基因+349位点A、G等位基因频率与肥胖有关。
Objective To investigate the relationship between adipon most abundant gene transcript1 +45 locus and +349 locus single nucleotide polymorphism and obesity. Methods The genotypes of +45 and +349 loci of adiponectin gene were determined by using optimized specific primer PCR (SSP-PCR). Results The frequencies of GT genotype at +45 locus in adiposity group and control group were higher than those in Uygur nationality. The frequencies of TT genotype in Uighur group were higher than those in Han nationality, but the difference was not statistically significant (P> 0.05) The frequency distribution of A and G alleles of +349 locus in adiposity group and control group of Uygur population was statistically significant (P <0.05). Conclusion There may be a difference in the +45 locus of adiponectin gene between Uygur and Han ethnic groups. The frequencies of A and G alleles at +349 locus in adiponectin gene in Uighur population are related to obesity.
