目的研究伊马替尼(imatinib)对Jurkat T细胞中A20、A20结合的核因子κB抑制物1(ABIN1)表达的影响。方法用(25、50、100)nmol/L imatinib处理Jurkat T细胞24 h,应用实时定量PCR检测A20、ABIN1、核因子κB(NF-κB)的mRNA水平,Western blot法检测A20、ABIN1、NF-κB蛋白水平。结果佛波酯/离子霉素混合物刺激Jurkat T细胞,可明显上调ABIN1和A20的mRNA和蛋白的表达。Imatinib明显抑制Jurkat T细胞ABIN1、NF-κB mRNA和蛋白的水平,上调A20 mRNA和蛋白水平。结论 Imatinib通过上调A20水平抑制NF-κB通路激活,该作用不需要ABIN1参与。
Objective To investigate the effect of imatinib on the expression of A20 and A20-bound nuclear factor κB inhibitor 1 (ABIN1) in Jurkat T cells. Methods Jurkat T cells were treated with (25, 50, 100) nmol / L imatinib for 24 hours. The mRNA levels of A20, ABIN1 and NF-κB were detected by real- -κB protein level. Results Phorbol ester / ionomycin mixture stimulated Jurkat T cells, can significantly up-regulate ABIN1 and A20 mRNA and protein expression. Imatinib significantly inhibited ABIN1, NF-κB mRNA and protein levels and up-regulated A20 mRNA and protein levels in Jurkat T cells. Conclusion Imatinib inhibits the activation of NF-κB pathway by up-regulating the A20 level, which does not require the participation of ABIN1.
