以优良玉米自交系H99与齐319为研究材料,采用正交试验设计L9(34),探讨了2,4-D、CaCl2、AgNO3三个因素不同浓度组合对玉米幼胚愈伤组织形成的影响。结果表明:2,4-D、CaCl2、Ag-NO3对两个自交系胚愈伤组织诱导率均有影响,其中H99所需2,4-D、CaCl2、AgNO3最佳浓度组合为2mg/L、5mmol/L、8mg/L或2mg/L、5mmol/L、12mg/L;自交系齐319所需2,4-D、CaCl2、AgNO3的最佳浓度组合为2mg/L、4mmol/L、10mg/L或2mg/L、5mmol/L、10mg/L。本研究为基于这两个自交系的玉米遗传转化提供了优化的愈伤组织培养条件。
The optimal maize inbred lines H99 and Qi 319 were used as materials to study the formation of immature embryos callus by orthogonal design L9 (34). The effects of 2,4-D, CaCl2 and AgNO3 influences. The results showed that 2,4-D, CaCl2 and Ag-NO3 had an effect on the induction rate of embryogenic calli in two inbred lines, in which the optimum concentrations of 2,4-D, CaCl2 and AgNO3 for H99 were 2mg / L, 5mmol / L, 8mg / L or 2mg / L, 5mmol / L and 12mg / L respectively. The optimal concentration of 2,4-D, CaCl2 and AgNO3 for inbred line Qi 319 was 2mg / L, 10 mg / L or 2 mg / L, 5 mmol / L, 10 mg / L. The present study provides optimized conditions for callus culture based on genetic transformation of maize from both inbred lines.
