目的研究A-549肺腺癌细胞株生长抑素受体亚型2、5(SSTR-2、SSTR-5)的表达情况及奥曲肽对A-549肺腺癌细胞株分泌转化生长因子β1(TGF-β1)的影响。方法采用S-P免疫组织化学法检测A-549细胞株中SSTR-2、SSTR-5的表达情况;采用酶联免疫吸附实验的方法(ELIZA)检测奥曲肽对A-549细胞分泌TGF-β1的作用。结果在A-549肺腺癌细胞株中,SSTR-2、SSTR-5均呈阳性表达。高浓度奥曲肽可以抑制A-549细胞自分泌的TGF-β1,且呈时间-剂量依赖性。结论 SSTR-2、SSTR-5于A-549肺腺癌细胞皆有表达;奥曲肽可以抑制A-549细胞分泌TGF-β1。
Objective To investigate the expression of somatostatin receptor subtype 2,5 (SSTR-2, SSTR-5) in A-549 lung adenocarcinoma cell lines and the effect of octreotide on the secretion of transforming growth factor β1 -β1). Methods S-P immunohistochemistry was used to detect the expression of SSTR-2 and SSTR-5 in A-549 cells. The effect of octreotide on the secretion of TGF-β1 by AQUA was detected by enzyme-linked immunosorbent assay (ELIZA) Results The positive expression of SSTR-2 and SSTR-5 was observed in A-549 lung adenocarcinoma cell lines. High concentration of octreotide can inhibit the autocrine TGF-β1 of A-549 cells in a time-and dose-dependent manner. Conclusions SSTR-2 and SSTR-5 are expressed in A-549 lung adenocarcinoma cells. Octreotide can inhibit the secretion of TGF-β1 by A-549 cells.
