目的对MSCs衰老的特征及发生机制的研究进展进行综述。方法广泛查阅近年来国内外研究MSCs衰老的相关文献,并进行分析总结。结果 MSCs在实验培养传代过程中存在着衰老迹象,表现为形态的衰老变化,增殖、分化能力逐渐减低及免疫调节功能的改变等,影响了其在临床的应用前景。目前关于MSCs衰老机制的研究主要集中在端粒与端粒酶调控、应激损伤等,涉及到端粒酶活性调控机制、p53/p21、P13K/Akt及Wnt/β-连环蛋白等信号通路的调节。结论对MSCs衰老机制的研究有助于加速MSCs向临床应用的转化。
Objective To review the characteristics and mechanism of MSCs aging. Methods Extensively review the literatures about the senescence of MSCs both at home and abroad in recent years and analyze and summarize. Results MSCs showed signs of aging in the process of passage of experimental culture, showing the change of aging, the gradual decrease of proliferation and differentiation and the change of immunomodulatory function, which affected the clinical application of MSCs. At present, the studies on the mechanism of MSCs aging mainly focus on the regulation of telomere and telomerase, stress injury and so on, which involve in the regulation of telomerase activity, p53 / p21, P13K / Akt and Wnt / β-catenin signaling pathways adjust. Conclusion The study of MSCs aging mechanism helps to accelerate the transformation of MSCs to clinical application.
