目的:观察川芎含药血清对体外培养的雪旺氏细胞(SCs)增殖及LNGFR、BDNF和CNTF表达的影响,探讨其促进周围神经再生的可能机制。方法:SD大鼠随机分为川芎组和空白组。川芎组分别灌胃不同剂量(1.25、2.5、5.0、10.0 g/kg)川芎水煎液,空白组给予等体积蒸馏水,1 w后经股动脉采血制备血清。选取出生3 d的SD乳大鼠双侧坐骨神经制备SCs,将纯化的SCs分为川芎含药血清组和空白血清组,MTT法测定不同时间各组光密度D(λ)值;48 h后应用RT-PCR法检测LNGFR、BDNF和CNTF mRNA表达水平。结果:川芎2.5、5.0、10.0 g/kg组血清培养的SCs在培养1 d后D(λ)值均大于空白血清组(P<0.05或P<0.01);与空白血清组比较,川芎含药血清组LNGFR mRNA表达显著升高(P<0.01)、BDNF和CNTF mRNA表达显著降低(P<0.01)。结论:2.5~10.0 g/kg川芎含药血清可有效促进新生大鼠SCs增殖,其机制可能与上调LNGFR和下调BDNF、CNTF mRNA的表达有关。
OBJECTIVE: To observe the effect of Chuanxiong rhizome serum on the proliferation of cultured Schwann cells (SCs) and the expression of LNGFR, BDNF and CNTF in vitro, and to explore the possible mechanism of the effect of Chuanxiong containing serum on promoting peripheral nerve regeneration. Methods: SD rats were randomly divided into Chuanxiong group and blank group. Chuanxiong group were given different doses (1.25,2.5,5.0,10.0 g / kg) of Chuanxiong decoction, the blank group were given an equal volume of distilled water, 1 w after the femoral artery blood serum preparation. SCs were prepared from bilateral sciatic nerves of SD rats born 3 days after birth. The purified SCs were divided into Chuanxiong group and blank serum group. The optical density D (λ) values of different groups at different time were determined by MTT method. The expression of LNGFR, BDNF and CNTF mRNA was detected by RT-PCR. Results: The D (λ) values of SCs incubated with 2.5, 5.0 and 10.0 g / kg of Chuanxiong rhizome were higher than that of blank serum group (P <0.05 or P <0.01), respectively. Compared with blank group, Serum LNGFR mRNA expression was significantly increased (P <0.01), BDNF and CNTF mRNA expression was significantly reduced (P <0.01). CONCLUSION: The serum of Chuanxiong 2.5 ~ 10.0 g / kg can effectively promote the proliferation of SCs in neonatal rats. The mechanism may be related to the up-regulation of LNGFR and down-regulation of the expression of BDNF and CNTF mRNA.
