目的优选复方榆树中咖啡酸的提取工艺。方法采用HPLC测定复方榆树中指标成分咖啡酸转移率,流动相:甲醇-0.4%磷酸(27∶73);色谱柱:Agilent Eclipse Plus C_(18)(4.6μm×250μm,5μm);柱温:30℃;检测波长:323 nm;检测器:二极管阵列检测器(DAD);分析时间:25 min;流速:1.0 m L·min~(-1);进样量:10μL。以咖啡酸的转移率为指标,通过单因素试验考察提取方法、料液比、提取时间、提取次数、粉碎度对提取工艺的影响,并采用正交试验优化提取工艺。结果最佳提取工艺为复方药材过60目筛网,用8倍量60%乙醇加热回流提取3次,每次30 min。结论优选的提取工艺稳定可行,为复方榆树的开发研究提供参考。
Objective To optimize the extraction process of caffeic acid in compound elm. Methods HPLC method was used to determine the caffeic acid transfer rate in the elm tree. The mobile phase consisted of methanol-0.4% phosphoric acid (27:73). The column was Agilent Eclipse Plus C18 (4.6μm × 250μm, 5μm) 30 ℃; detection wavelength: 323 nm; detector: diode array detector (DAD); analysis time: 25 min; flow rate: 1.0 m L · min -1; injection volume: 10 μL. Taking the caffeic acid transfer rate as an index, the effects of extraction method, solid-liquid ratio, extraction time, extraction times and degree of pulverization on the extraction process were investigated by single factor experiments. The orthogonal experiment was used to optimize the extraction process. Results The best extraction process for the compound herbs through 60 mesh sieve, with 8 times the amount of 60% ethanol reflux heating extraction three times, each time 30 min. Conclusion The optimal extraction process is stable and feasible, providing a reference for the development of compound elm.
