[目的]为探索MS2噬菌体的宿主菌范围及在不同的宿主菌内的生长情况等,为流行病学研究、细菌的噬菌体分类鉴定和其他相关研究提供基本数据和资料。[方法]采用双层琼脂法测定大肠埃希菌MS2噬菌体对各试验菌株的裂解情况,观察其噬菌斑生长情况并计数。[结果]①大肠埃希菌MS2噬菌体与埃希大肠杆菌(ATCC15597)、大肠杆菌285和大肠杆菌B分别进行双层琼脂平板培养,结果均出现大小均一、边缘整齐、透明清晰的噬菌斑;②相同稀释度的MS2噬菌体与埃希大肠杆菌(ATCC15597)、大肠杆菌285和大肠杆菌B培养产生的噬菌斑数量依次递减;③MS2噬菌体与其余6种所选试验菌株分别进行双层琼脂平板培养,结果琼脂表面均出现浑浊,未产生噬菌斑。[结论]在本次试验所选的8种试验菌中,通过双层琼脂法测定适合做大肠埃希菌MS2噬菌体宿主菌者有埃希大肠杆菌(ATCC15597)、大肠杆菌285和大肠杆菌B;所选其余6种试验菌株均不适合做MS2噬菌体的宿主菌。
[Objective] To provide basic data and information for epidemiological studies, bacteriophage classification and identification of bacteria and other related research in order to explore the range of host bacteria of MS2 phage and its growth in different host bacteria. [Method] The double-layer agar assay was used to determine the cleavage of Escherichia coli MS2 phage against each experimental strain. The plaque growth was observed and counted. [Results] (1) Escherichia coli MS2 phage and Escherichia coli (ATCC15597), Escherichia coli 285 and Escherichia coli B were double-layer agar plate culture, the results were uniform size, neat edge, clear and transparent plaques; ② The same dilution of MS2 phage and Escherichia coli (ATCC15597), Escherichia coli 285 and Escherichia coli B plagued the number of plaques decreased; ③ MS2 phage and the remaining six selected strains were double-agar plate culture As a result, the surface of the agar appeared cloudy, did not produce plaques. [Conclusion] Among the 8 kinds of test bacteria selected in this experiment, Escherichia coli (ATCC15597), Escherichia coli 285 and Escherichia coli B were identified by double-layer agar assay for the host strains of Escherichia coli MS2 phage. The other six selected test strains are not suitable for MS2 phage host bacteria.
