目的探讨甲状腺激素(TH)对新生大鼠脑血管发育的影响。方法取18只清洁级SD成年雌性大鼠与3只清洁级SD雄性大鼠交配后随机将孕鼠分入3组:丙基硫氧嘧啶(PTU)干预组、TH干预组和对照组。待每组仔鼠产后第1天即采用电化学发光法检测其血清总三碘甲状腺原氨酸(TT3)、总甲状腺素(TT4)水平;采用酶联免疫吸附试验法测仔鼠脑组织血管内皮细胞生长因子(VEGF)水平。应用SPSS 16.0软件进行统计学分析。结果 TH干预组仔鼠血清TT4[(103.50±2.37)μg·L-1]、TT3[(1269.63±32.18)ng·L-1]与脑组织VEGF水平[(53.46±10.62)μg·L-1]均高于对照组[TT4(45.11±0.43)μg·L-1,TT3(844.57±20.85)ng·L-1,VEGF(11.15±1.33)μg·L-1](Pa<0.001)。同时,PTU干预组新生鼠以上各指标[TT4(16.83±0.45)μg·L-1,TT3(142.35±7.76)ng·L-1,VEGF(8.69±0.48)μg·L-1]均低于对照组(Pa<0.001)。结论 TH干预可增加仔鼠脑组织VEGF水平,对脑血管的发育有促进作用。
Objective To investigate the effect of thyroid hormone (TH) on cerebrovascular development in neonatal rats. Methods Eighteen SD female SD rats were randomly assigned to three groups: PTU intervention group, TH intervention group and control group. Each group of pups were sacrificed on the first day of postpartum, and their serum total triiodothyronine (TT3) and total thyroxine (TT4) levels were measured by electrochemiluminescence method. The blood vessels of the brains of the pups were measured by enzyme linked immunosorbent assay Endothelial cell growth factor (VEGF) levels. Application SPSS 16.0 software for statistical analysis. Results The levels of TT4 [(103.50 ± 2.37) μg · L-1], TT3 [(1269.63 ± 32.18) ng · L-1] and the levels of VEGF in brain tissue of TH intervention group [(53.46 ± 10.62) μg · L- ] Were higher than those in the control group [TT4 (45.11 ± 0.43) μg · L-1, TT3 (844.57 ± 20.85) ng · L-1, VEGF (11.15 ± 1.33) μg · L-1] At the same time, each index of the newborn rats in the PTU intervention group was significantly lower than those in TT4 (16.83 ± 0.45) μg · L-1, TT3 (142.35 ± 7.76) ng · L-1 and VEGF (8.69 ± 0.48) μg · L-1 Control group (Pa <0.001). Conclusions TH intervention can increase the level of VEGF in the brain tissue of the offspring and promote the development of cerebrovascular.
